Extraction and Purification Of Alcohol Dehydrogenase From Baker’s Yeast (Saccharomyces Cerevisiae)

Abstract:
The purification of alcohol dehydrogenase (ADH) from Baker’s Yeast (Saccharomyces Cerevisiae) was determined by investigation into several different purification techniques. In this work aqueous two phase extract (ATPE) and ammonium sulfate precipitate were examined to determine the most effective initial purification technique. Several variants of ATPE were examined in this work followed by gel filtration. Enzyme activity was monitored at each purification step, resulting with the highest enzyme activities being found in the bottom layer of the ATPE systems, with the ATPE makeup of 15% polyethylene glycol (PEG) and 40% ammonium sulfate having the highest specific activity.
